Depends on what your interests are.
If it's a family with trip with young kids, The Franklin Institute is a must.
If you are into more highbrow culture The Philadelphia Museum of Art would be a great place to visit.
If you are into live music, Union Transfer is a cool venue to see some great original music.
If you are looking for Philly flavored eats, try the Reading Terminal Market, there is something for everyone there. - BiggE

If you are the active sort, i'd add if you like hikes check out Fairmount park/Wissahickon. if you want to bike - get a bike at boathouse row and ride on kelly drive, and/or the tow path in Manayunk and/or the forbidden drive in Wissahickon. - nastyflyergirl
